Description: HyFusion™ Hybridoma Fusion Reagent, balanced 50% PEG, is a proprietary fusion reagent for fusion of myeloma cells with spleen cells. It is based on PEG1500 with added buffer and stabilizers for highest fusion efficiency and stability of cells. Use this reagent if you are doing fusion to generate new hyrbidomas. This is cell culture tested and guaranteed to give high fusion rate. This is provided as a sterile and ready to use solution in a buffered and balanced media. Detailed protocol on how to use this product is available on our website.

Usage: Pool spleen cells with myeloma cells in appropriate ratio (see our detailed protocol for best results) in a 50mL centrifuge tube. Centrifuge at 1000rpm for 5 minutes. Discard supernatant. Drain completely of all liquid and then resuspend pellet by gentle tapping. Add 1mL of HyFusion Balanced 50% PEG Hybridoma Fusion Reagent, per 100 million spleen cells used, to the tube dropwise on the side of wall with slow rotation in a 37C water bath. Continue rotating the tube for 2 minutes, then add 1mL of media without sera. Incubate further 2 minutes by gentle swirling in 37C water bath. Then fill the tube with media without sera to 50mL, centrifuge and continue with rest of the steps. It is recommended to use our HyMax™ Hybridoma Fusion and Growth Supplement (Item# 113004) for culturing freshly fused cells without feeder cells (check our website for more information). You may also use our Ag8 myeloma cell line (Item#116036) for fusion with your immunized mouse or rat spleen cells

Format: Provided as a sterile filtered, tissue culture tested, ready to use solution. Never frozen. Each package comes with 50mL reagent enough to do 25 to 50 fusions.

Storage: Store at 4°C - 8°C in refrigerator until use. Do not freeze.

Stability: 18 months when stored at 4°C - 8°C in refrigerator as recommended.
